The Effect of JunD Knockdown on Gene Expression and Protein Levels in Prostate Cancer Cells

Abstract Summary

Prostate cancer (PCa) is one of the most frequently diagnosed malignancies among men worldwide and the second leading cause of cancer-related deaths in the United States. In many cases, PCa develops slowly, but it can be more aggressive and metastasize to other parts of the body. Our recent studies demonstrate that JunD plays an essential role in the proliferation of PCa cells and may provide a significant therapeutic target for the treatment of PCa. In this study we focused on genes that may be regulated by JunD. Selective knock-down (KD) of JunD using siRNA in DU145 and PC3 cells was performed followed by Proteomic and Microarray analysis. We selected the top down-regulated genes and analyzed their gene and protein levels by qRT-PCR and Western blot analysis, respectively. Our results show that the JunD-KD decreases both mRNA and protein levels of known and novel genes involved in cell proliferation. Future studies will include further analysis of these selected genes and their effects on cell proliferation in PCa cell lines.
